After seeing the promo, I can't wait to see the movie. Spidey got me back into reading comics after I had quit when they went up to $.25 way back when, so I have a particularly soft spot in my heart for him.
I loved what they did with Doc Ock - any writer who can make him a sympathetic character has some real writing skills, in my opinion. I'm hoping they can do the same thing with Sandman, whom I have always considered a pretty pathetic villain. I'm sorry to see that they're going to make him Uncle Ben's killer though. The whole Spider-Man schtick is that it was through Peter's lack of responsibility that UB was murdered. If it wasn't, then there's really no point, hmm?
I'm also hoping that they don't run into the latter Batman movie error of overwhelming the plot with too many villains. My personal expectation after SM2 was that we would see Harry come back as the new Green Goblin, or else Dr. Connor going to the bad.
All that being said, I'll be plunking down my $6.50 (bargan matinee) when it hits the big screen.

But Marvel does have the best explination for why instead of dieing from radiation or gene altering serums. Long ago the Celestials altered the human genome and added a certain seed that can either be activated naturally (mutants) or from some trauma (everyone else).

Comics very often try to provide a scientific explanation of the powers of super-heroes as well -- take a look at the early X-Men for one example among many. It's just that they do it in a small footnote and jump straight to the action while a two-hour movie needs a more detailed description.
